SRX25775146: Whole genome sequence of Bacillus_thuringiensis_SC136

Design: individual strains were isolated, following 10 minutes incubation at 95 degrees, by overnight growth at 37 degrees on TSA+5% blood agar, and were colony purified. 5 ml Tryptic soy broth cultures were grown for approximately 4 hours at 37 degrees C, cells were harvested by centrifugation, and DNA was isolated using a Promega wizard kit following digestion with lysozyme. Libraries were prepared and sequenced as 151 bp paired end reads using an Illumina NovaSeq X Plus by SeqCenter, who prepared libraries using the illumina DNA prep kit and custom IDT 10bp unique duel indices.

Biology students at Siena College isolate, characterize, and genome sequence endospore forming soil bacteria from campus soil. We intentionally select strains with less common colony types during the isolation process, to increase data about under-sampled Bacillaceae.
